IgE mediates broncho-vascular remodeling after neonatal sensitization in mice.

چکیده
The temporal origins of childhood asthma are incompletely understood. We hypothesize that allergen sensitization which begins in early infancy causes IgE-mediated airway and vascular remodeling, and airway hyper-responsiveness. Mice were sensitized with ovalbumin (OVA) without or with anti-IgE antibody from postnatal day (P) 10 through P42. We studied airway resistance in response to Methacholine (MCh) challenge, bronchoalveolar lavage fluid (BAL) inflammatory cell content, immunohistochemistry for inflammation, alpha-smooth muscle actin (alpha-SMA) and platelet/endothelial cell adhesion molecule (PECAM) proteins, and Western blotting for vascular endothelial growth factor (VEGF) protein. Compared to controls, mice treated with OVA had increased airway resistance (baseline: 192% of control; MCH 12 mg/mL 170% of control; P less than 0.0.5). OVA treatment also increased lung alpha-SMA, VEGF and PECAM compared to controls. Inflammatory cells in the BAL and perivascular and peribronchiolar inflammatory cell infiltrates increased over controls with OVA exposure. These changes were counteracted by anti-IgE treatment. We conclude that mice sensitized in early infancy develop an IgE-mediated hyper-reactive airway disease with airway and vascular remodeling. Preventive approaches in early infancy of at-risk individuals may reduce childhood asthma.

NFATc3 is required for chronic hypoxia-induced pulmonary hypertension in adult and neonatal mice.
Pulmonary hypertension occurs with prolonged exposure to chronic hypoxia in both adults and neonates. The Ca(2+)-dependent transcription factor, nuclear factor of activated T cells isoform c3 (NFATc3), has been implicated in chronic hypoxia-induced pulmonary arterial remodeling in adult mice. Therefore, we hypothesized that NFATc3 is required for chronic hypoxia-induced pulmonary hypertension i...
متن کاملT Helper 2 Cytokine Analysis of Bronchoalveolar Lavage in the Murine Model of Allergic Broncho Pulmonary Aspergillosis
Background and Aims: Aspergillus fumigatus is a sporadic fungus that causes different infections and allergies in immunocompromised patients. The allergic disease caused by this fungus is called allergic bronchopulmonary aspergillosis (ABPA). ABPA is considred important in atopic and immunocompromised individuals, which can result in inflammation and epithelial damage. Therefore, the aim of thi...
متن کاملMaternal Bisphenol A Exposure Promotes the Development of Experimental Asthma in Mouse Pups
BACKGROUND We recently reported that various environmental estrogens induce mast cell degranulation and enhance IgE-mediated release of allergic mediators in vitro. OBJECTIVES We hypothesized that environmental estrogens would enhance allergic sensitization as well as bronchial inflammation and responsiveness. To test this hypothesis, we exposed fetal and neonatal mice to the common environme...
متن کاملSensitization by subcutaneous route is superior to intraperitoneal route in induction of asthma by house dust mite in a murine mode
OBJECTIVE To develop a new experimental model of chronic allergic pulmonary disease induced by house dust mite, with marked production of specific immunoglobulin E (IgE), eosinophilic inflammatory infiltrate in the airways and remodeling, comparing two different routes of sensitization. METHODS The protocol lasted 30 days. BALB/c mice were divided into six groups and were sensitized subcutane...
متن کاملAntibody-antigen interaction in the airway drives early granulocyte recruitment through BLT1.
Antibody-antigen interactions in the airway initiate inflammation in acute asthma exacerbations. This inflammatory response is characterized by the recruitment of granulocytes into the airways. In murine models of asthma, granulocyte recruitment into the lung contributes to the development of airway hyperresponsiveness (AHR), mucus production, and airway remodeling. Leukotriene B4 is a mediator...
